Don Pedro Lake can be accessed from Highway 49 from Sonora on the north or Coulterville to the south. Highway 120, which leaves Highway 108 west of Sonora, is the most direct route to Don Pedro Lake from the Oakdale - Manteca area and north.

From Modesto, and most places south of Modesto, Highway 132 leads directly to Don Pedro Lake. County road J59 (La Grange Road) and Bond Flat Road run along the west side of the lake, connecting Highway 132 with Highway 120.
